漂白(bleaching)

手順

1. 排液する
2. オキシドールを0.2mlほど加える。
3. 10%アンモニア水を0.1mlほど加える。
4. 常温でしばらく置く(10分〜2時間程度)。
5. ときどき実体顕微鏡で観察して 適当な濃度になったら排液して氷酢酸を加える。

動画は2倍速。

注意事項

1. やりすぎると後戻りできないので慎重に行うこと。 もしやりすぎた場合は染色するしか無い。

考察

Brown2006より。

for specimens with opaque black cuticle, rinse in strong alcohol and then partially bleach cuticle by immersing in a freshly prepared mixture of cold strong Ammonia and 30-volume Hydrogen Peroxide solutions. Bleaching should be monitored as it can be very rapid, and may be stopped quickly by adding a few drops of a water-soluble acid. Domestic bleach is unsatisfactory for controlled cuticular bleaching.

かなり高濃度の薬品が使われているようなのだけど、 容易に入手可能なオキシドールと10％アンモニア水でできているので いいことにしよう。 時間がかかっているかも知れない。

家庭用の漂白剤は使えないようなことが書いてあるな。 酸素系もダメなのだろうか？
